The 2012 Broad Institute analysis is the study that made GHK-Cu interesting beyond cosmetics, and the useful part is the direction of the changes rather than the count. Collagen I rose roughly 70% in fibroblast models and collagen III around 50%, alongside elastin, glycosaminoglycans, superoxide dismutase and catalase. Moving the other way were inflammatory mediators, senescence markers and the matrix metalloproteinases that break structural protein down. Building the matrix while suppressing what degrades it is an unusual pairing. Endogenous plasma levels fall roughly 60% between age twenty and sixty, which is the argument for supplying it at all.
SNAP-8 Is Not Topical Botox
Both act on the SNARE complex, which is where the comparison should stop. Botulinum toxin cleaves SNARE proteins outright and produces complete neuromuscular blockade at the site. SNAP-8 is an eight-residue extension of the SNAP-25 N-terminus that competes for complex assembly, reducing contraction intensity rather than eliminating it. Preclinical in vitro work put wrinkle depth reduction at up to 63%, which is a real effect and a different order of magnitude from blockade. The practical consequences are that SNAP-8 is dose-titratable, reversible on a short timescale, and stacks with GHK-Cu because the two address unrelated causes.
Choosing Between Melanotan I and II
Melanotan I binds MC1R with relatively high selectivity, so its effect profile is narrow — melanogenesis and little else. Melanotan II is promiscuous across the melanocortin family, and the MC4R activity produces appetite suppression plus, in a substantial fraction of subjects, spontaneous erection as a well-documented off-target effect. Neither removes the need for caution around existing nevi, and both are typically run as a short loading phase followed by low-frequency maintenance rather than continuously. If pigmentation is the only endpoint, MT-I is the cleaner choice.
What the Blends Actually Contain
GLOW is GHK-Cu with co-factors aimed at the collagen pathway. KLOW extends that with additional peptides covering repair and expression-line mechanisms in the same vial.
